Description
The goals of this application are to develop human thalamic organoids (hThO), and to model human thalamocortical circuits in normal and neuropathological conditions such as schizophrenia (SZ) by fusing the hTO with human cortical organoids (hCO). In the proposed works, we will determine intrinsic electrophysiological properties of thalamic neurons in hThO using whole-cell patch-clamp recordings. Post hoc immunostaining of recorded cells following whole-cell patch-clamp recordings will be performed to determine if they express cellular markers for thalamocortical relay or inhibitory cells. We will measure synaptic activity and network oscillations using whole-cell patch-clamp recordings, local field potential recordings, and selective optogenetic activation of neuronal populations.
