(Aim 1.6 Approach 1) has been suppressed. In addition, following the reviewer's suggestion, we dropped the ectopic overexpression system proposed (previous Aim 1.8 Approach 2). A new Aim 1.6 ("Functional analysis of the interaction between the TSAd and Lck: regulation of Lck enzymatic activity") is included in the new proposal. It will add functional data to the structural requirements of the interaction between Lck and TSAd. We believe this is a significant addition to the original project that also responds to reviewer's (#2) suggestion ?...) to further investigate the already established mutations.... The fact that we can effectively perform experiments in primary TSAd-deficient T cells reconstituted with different modified forms of TSAd has been a major leap to assess structure/function relationship between TSAd and Lck.
