To define the role of endocytosis in regulating the activity of individual components of the MAPK pathway. Reagents necessary for further reconstgitution of the entire pathway will be developed. During the independent stage of this award, the MAPK pathway will be reconstituted in living cells. Time and sites of spatial overlap of individual components of the MAPK signaling cascade will be determined. We will also test whether the regulatory mechanisms of the spatiotemporal regulation of the components of the MAPK cascades are altered in cancer cells with different levels of EGFR.
