Abstract

A new sensitive method for quantitative determination of poly-3-hydroxybutyrate (PHB) and 3-hydroxybutyrate-3-hydroxyvalerate copolymer (PHBV) in microbial biomass by isocratic reversed-phase high-performance liquid chromatography was developed. The method employs chromatographic analysis of crotonic acid formed by PHB hydrolysis with concentrated sulfuric acid or crotonic and methylcrotonic acids formed by PHBV hydrolysis with 1 N NaOH.
