Abstract

After the publication of the Report "Generation of influenza A viruses as live but replication-incompetent virus vaccines," the authors were alerted to several issues in their supplementary figures, as described below. These figures have been corrected and are included in a revised version of the PDF supplement. Figure S4 presents the results of a screen designed to optimize sites for introducing premature termination codons (PTCs) into influenza A viruses. A high-throughput assay measuring the cytopathic effect (CPE) that measures cell damage during virus infection was used to select for viruses that had a significant CPE in the presence of a PTC, but no CPE in the absence of the PTC. Of 1760 panels collected, 426 were selected to show in Figure S4. Unfortunately, in compiling Figure S4A, several panels were either switched or duplicated. In Figure S15B there was unintentional duplication. To show that the unnatural amino acid Nε-2-azidoethyloxycarbonyl-L-lysine (NAEK) is nontoxic, the authors treated mice with NAEK either intramuscularly or intravenously. Heart tissues were stained with hematoxylin and eosin, and pictures were taken of three different areas (sites 1,2, and 3). When Figure S15B was generated, site 2 in the intravenous NEAK group was mistakenly used in the intramuscular NAEK panel instead of using site 1 in the intramuscular NAEK group. Images for intravenous and intramuscular administration of NAEK in the original figure have a duplicated region, because they both come from the same sample (intravenous NAEK group) and there is overlap between sites 1 and 2. The corrections made to Figure S4A, Figure S8A, and Figure S15B are described in detail, with before-and-after images (and, in the case of Figure S15B, raw data), at the end of the corrected supplement.
