N2 - The unusual dodecaketide premithramycinone G (see formula), which possesses a five-membered ring with two ketide chains in its tetracyclic structure, was accumulated upon inactivation of an early acting oxygenase of the mithramycin biosynthetic pathway. This unexpected structure allows novel hypothetic conclusions to be drawn regarding the early mithramycin pathway and suggestions for the involvement of three previously unassigned oxygenases. (Chemical Equation Presented).
