Abstract
The gene for the mouse prostaglandin 12 receptor (PGI2R) was isolated by screening a mouse PI genomic library using PCR. This screening was performed by Genomic Systems, Tnc, St. Louis, MO. The coding sequence of the gene is approximately 4 kbp is size. The receptor's coding sequence is interrupted by a single intron of about 1 kbp in size. This intron is located at +858 (A of ATG translation start site = +1). This is identical to the location of the intron found in the thromboxane A2 and PGÜ2 receptor genes. Unlike the human thromboxane A2 receptor gene, which contains an intron at -83 bp upstream from the translational start codon, no such intron was found in the mouse PGI2R gene. The sequence of the 266 bp of 5' region directly upstream from the coding sequence of the mouse receptor cDNA (Namba, et. al. (1994) J. Biol. Chem. 269, 998-9992) is identical to the corresponding upstream sequence in the mouse gene. However, since the transcriptional start site for the mouse PGI2R gene has not yet been determined, this does not rule out the possibility of the presence of an intron between the promoter and the translational start site.
