Synthesis of polymers for a gene carrier by the ester-amide exchange method

Abstract

Polyethylene glycol-polycation block copolymers, in which various kind of cationic side chains were introduced, were successfully synthesized by aminolysis of benzyl ester of polyethylene glycol-poly(β- benzyl-L-aspartate) block copolymer(PEG-PBLA)(ester-amide exchange method). Among them, some block copolymers showed the high transfection efficiency, and the mechanism and the characteristics of the ester-amide exchange reaction were investigated. This method is a very useful tool for design of non-viral gene carriers.
