Resumen
Chemical cross-linking strategies to determine protein conformation were discussed. Several model proteins such as myoglobin and cytochrome C were reacted with a combinatorial set of amine-specific cross-linking reagents. The set of crosslinkers with different spacers provided different maximum distance constraints between targeted residues. High performance liquid chromatography and mass spectrometry were used to confirm peptide cross-links and to investigate fragmentation pathways.
