Resumen

Quality sample preparation is an important prerequisite for downstream analysis of nucleic acids (NAs). Currently, this ubiquitous process requires a significant fraction of the available workload within many life science laboratories. In this manuscript, we expedite NA sample purification by utilizing an immiscible phase to filter contaminants (e.g. proteins, cell debris, lysis buffer) in a single step. In this process, paramagnetic particles (PMPs) which selectively bind NAs are magnetically driven across an immiscible barrier, termed the Phase-Gate. Aided by the surface tension-dominant physics associated with the microscale, this technique reduces purification time by ∼67% without sacrificing other aspects of performance.
